Materials

Refined soy lecithin Lipoid S100 was from Germany Lipoid GmbH (No.790495–1). Cholesterol was from Beijing Chemical Reagent Co. Quercetin was from Hunan nine modern Chinese Medicine Co. The hematoxylin and eosin (H&E) staining kit was from Shanghai Huyu Biological Technology Co. GPT/GOT ELISA kit was from Shanghai Ximei Biological Technology Co. Bilirubin assay kit was from Shanghai Hengyuan Biological Technology Co. Carbon tetrachloride was from Beijing Jinhuitaiya Chemical Reagent Co. All other reagents such as anhydrous alcohol,ether, and methanol were of analytical grade.

Rats were sacrificed by taking femoral artery blood after 1 h, 3 h, 6 h and 24 h of reperfusion, respectively. The liver tissue was taken and frozen at –20°C in a low temperature refrigerator. The tissue was thawed in a mixture of ice and water and prepared as 10% homogenate. The SOD and MDA were determined according to the instructions of kits from Shanghai Hengyuan, China.

2.5. Determination of serum testosterone

We evaluated serum testosterone level in rats by ELISA. After the rats were sacrificed, approximately 5 mL of abdominal aortic blood was collected and subjected to centrifugation at 3000 rpm for 15 min, and the serum was carefully aspirated and stored at ? 20 ?C for later use. According to the instructions for the rat testosterone ELISA kit (Hengyuan Biotech, Shanghai, China), we used different concentrations of testosterone standards and corresponding absorbance values to prepare a standard curve, and thus determined the testosterone in each group of samples after observing the sample absorbance.
