The whole buccal and palatal periodontal tissues of the upper molars were collected and homogenized in PBS. The samples were centrifuged for 15 min at 5000 rpm and 4 ?C. The supernatants were stored at ? 80 ?C for further analysis to evaluate the levels of interleukin 1β (IL-1β), IL-6, IL-8, tumor necrosis factor-α (TNF-α) and prostaglandin E2 (PGE2). The cytokine levels were measured by ELISA assay following the manufacturer’s instructions. The absorbance was measured at 450 nm. Results were calculated using the standard curves created in each assay and shown as ng/L.

To evaluate the effect of the implanted scaffold on local skin tissues, the skins surrounding the hydrogels were collected to examine the inflammatory cell infiltration and tissue damage via hematoxylin and eosin (H&E) staining on day 4. The levels of inflammatory cytokines (IL- 1, IL-6 and TNF-α) within the tumor tissues were also measured using ELISA methods.

Microbiological medium: Lysogeny Broth (LB), Nutrient Agar (NA), Potato Dextrose Agar (PDA), Man Rogosa Sharpe (MRS) broth, Violet Red Bile Agar (VRBA) medium were purchased from Shanghai Hengyuan Biotechnology Co., ltd (Shanghai, China). Standard chemicals: rutin; gallic acid were bought from Tianjin Hengxing Chemical Reagent Manufacturing Co. ltd (Tianjing, China). Lactic acid bacteria strain SH- 470 consisting of Lactiplantibacillus Plantarum (STCC0007) and Streptococcus thermophilus (STCC0038) was bought from Shenghe Biotechnology Co., ltd (Yangzhou, China). Staphylococcus aureus (S. aureus, ATCC6538); Escherichia coli O157:H7 (E. coli O157:H7, NCTC12900); Aspergillus flavus spores (A. flavus, CGMCC3.3950) and Saccharomyces cerevisiae (S. cerevisiae, ATCC9763) were obtained from Beijing Microbiology Technology Co., ltd (Beijing, China).
